Abstract

Ceramides are critical components of human skin,with their structural composition—particularly the distribution of fatty acid chain lengths—playing a key role in determining their functional properties.The fatty acid chain length distribution in plant oils closely resembles that of ceramides in the stratum corneum.In this study,ceramide NP with fatty acid chains ranging from C16 to C24 was synthesized using Longnan extra virgin olive oil as a fatty acid donor.Transcriptomic analysis was employed to evaluate the protective effects of olive oil-derived ceramide NP on the skin barrier under UVB exposure,including enrichment analysis of differentially expressed genes.In addition,an SLS-induced epidermal skin model was used to assess impacts on inflammatory factor expression,tissue morphology,and levels of barrier-related proteins.Results indicated that olive oil-derived ceramide NP exerted soothing and reparative effects by inhibiting inflammation-related signaling pathways,reducing inflammatory mediator secretion,maintaining tissue integrity,and upregulating barrier-associated proteins.Compared with stearic acid-derived ceramides(which contain a single fatty acid chain length),olive oil-derived ceramide NP showed significantly enhanced efficacy at equivalent concentrations.
