'金如意'山楂叶绿体基因组特征与系统进化分析
Characterization and Phylogenetic Analysis of Complete Chloroplast Genome of Crataegus pinnatifida Bge.var.major Jinruyi

Abstract
Using the leaves of Crataegus pinnatifida Bge.var.major Jinruyi as materials,the complete chloroplast genome sequence was sequenced,assembled and analyzed by Illumina high-throughput sequencing technology in this study.The phylogenetic analysis was also conducted on the chloroplast genomes of 34 Rosa-ceae species.The results showed that the complete chloroplast genome of C.pinnatifida Bge.var.major Jinruyi was 159 655 bp in length with the GC content of 36.65%,and contained a large single copy(LSC)region of 87 748 bp,a small single copy(SSC)region of 19 139 bp and a pair of inverted repeats(IR)regions of 26 384 bp.A total of 130 genes were encoded,including 85 protein-coding genes,37 tRNA genes,and 8 rRNA genes.A total of 264 SSR loci and 57 repeats were detected by repeat analysis with most SSR loci con-sisting of A-and T-repeat bases.There were 31 high-frequency codons(RSCU>1),29 of which ended with A/U.Compared with other Rosaceae species,the IR region boundary of the C.pinnatifida Bge.var.major Jin-ruyi chloroplast genome was relatively conserved.The phylogenetic analysis showed that C.pinnatifida Bge.var.major Jinruyi was most closely related to C.pinnatifida var.major(GenBank No.NC_065486.1),indica-ting that it belonged to the large-fruit hawhorn type.These findings could lay a foundation for the identification of hawthorn germplasm resources,the analysis of genetic diversity,and the utilization of hawthorn resources.关键词
