Quality analysis of grafted Ziziphi Spinosae Semen based on DNA barcode and UPLC-MS/MS technology

Abstract
Objective The quality of grafted Ziziphi Spinosae Semen was analyzed by DNA barcode technology,UPLC-MS/MS technology and chemometrics.Methods The 18 batches of grafted Ziziphi Spinosae Semen,wild Ziziphi Spinosae Semen,and wild artificially cultivated Ziziphi Spinosae Semen were collected for DNA extraction,PCR amplification,agarose gel electrophoresis and bidirectional sequence.The sequences were compared by DNAMAN analysis software.The content of 10 characteristic components in 18 batches of grafted Ziziphi Spinosae Semen was measured using UPLC-MS/MS technology.The content determination results were comprehensively analyzed using hierachical clustering analysis(HCA),principal component analysis(PCA),and orthogonal partial least-squares discrimination analysis(OPLS-DA).Results The 18 batches of grafted Ziziphi Spinosae Semen were essentially the same as the wild Ziziphi Spinosae Semen in terms of basic origin,and the number of grafting years had minimal effect on the contents of 10 characteristic components in the grafted Ziziphi Spinosae Semen.Jujuboside B,jujuboside A,aconitine,spinosin and isovitexin were representative differential components in grafted Ziziphi Spinosae Semen,which served as differential markers affecting the quality of the grafted Ziziphi Spinosae Semen.Conclusion The quality analysis of grafted Ziziphi Spinosae Semen,focusing on aspects of basic origin and characteristic component content determination,provided a theoretical basis for the effectiveness in clinical use of grafted Ziziphi Spinosae Semen.关键词
