Preventive and Therapeutic Effects of Dapagliflozin on Gouty Arthritis in Rats Based on Network Pharmacology-Molecular Docking and Animal Experiments

Abstract

In order to explore the preventive and therapeutic effects of dapagliflozin(DAPA)on gouty arthri-tis(GA)in rats,the signaling pathways of DAPA in the treatment of GA were screened using network pharma-cology and molecular docking technology,and the GA model of SD rats was constructed by injecting sodium urate crystals into the ankle.The swelling degree of the ankle was measured by a vernier caliper,and the pathological changes of the ankle were observed by hematoxylin and eosin staining.The levels of inflamma-tory factors were detected by enzyme-linked immunosorbent assay,and the expression of nucleotide binding oligomerization domain(NOD)-like receptor family pyrin domain containing 3(NLRP3)and caspase-1 in the synovial tissues of the ankle joint was detected by Western-blot.The results of network pharmacology and molecular docking showed that the potential targets of DAPA in the treatment of GA were significantly en-riched in the NOD-like receptor signaling pathway(P<0.01),and the binding energies of all core targets in this pathway to DAPA were lower than-29.29 kJ/mol.Animal experiments showed that DAPA significantly reduced ankle swelling,ameliorated pathological injury,decreased serum levels of inflammatory cytokines interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α),and suppressed the expression of NLRP3 and caspase-1 in synovial tissues of GA rats.These results suggest that DAPA may alleviate acute inflammatory responses of GA rats by inhibiting the NLRP3/caspase-1 signaling pathway.
