摘要

Abstract

As an infectious viral pathogen,human metapneumovirus(hMPV)can cause severe respiratory in-fections and is classified as one of the important pathogens by the World Health Organization(WHO).Al-though vaccines play an important role in disease prevention and transmission control,the wild-type viruses are prone to immune escape due to the relatively high mutation rates of their structural proteins.Therefore,developing broad-spectrum hMPV vaccines is essential to provide extensive protection against multiple viral variants.Herein,based on consensus sequence methodology,a broad-spectrum T-cell epitope vaccine was designed,which was composed of 478 amino acid residues,containing 10 cytotoxic T lymphocyte(CTL)epi-topes and 11 helper T lymphocyte(HTL)epitopes from the viral fusion protein(F),attachment glycoprotein(G),matrix protein(M),and small hydrophobic protein(SH).The designed multi-epitope vaccine demonstrated favorable characteristics:no allergenicity,high population coverage,strong antigenicity and immunogenicity,appropriate physicochemical properties,and high solubility.Meanwhile,the vaccine structure exhibited high similarity to that of the natural virus.Structural biology analysis revealed that the engineered vaccine achieved superior performance in both structural compactness and binding stability.Computational immunology simu-lations demonstrated that the vaccine could elicit immune responses in humans.In conclusion,the broad-spectrum hMPV vaccine developed in this study can be a promising candidate for preventing hMPV infec-tion,and the prediction pipeline established here proved an efficient method for screening immunodominant epitopes of other pathogens.
