Abstract
Objective To investigate the effect of microRNA-154(miR-154)on Wnt/β-catenin signaling pathway in regulating collagen expression in angiotensin Ⅱ(Ang Ⅱ)-induced cardiac fibroblasts.Methods Cardiac fibroblasts were isolated and cultured from neonatal rats,followed by Ang Ⅱ treatment.miR-154 expression was detected by quantitative real-time PCR(qRT-PCR).Subsequently,miR-154 inhibitor was transfected into cardiac fibroblasts prior to Ang Ⅱ stimulation.The down-regulation effect was verified by qRT-PCR.Western blot was used to detect protein expression levels of β-catenin,c-myc,collagen typeⅠ(CoL-Ⅰ),collagen type Ⅲ(CoL-Ⅲ),and α-smooth muscle actin(α-SMA)in cells.Cardiac fibroblasts transfected with miR-154 inhibitor were treated with Wnt/β-catenin activator.The expressions of β-catenin,c-myc,CoL-Ⅰ,CoL-Ⅲ,and α-SMA were determined by Western blot.Results The expression of miR-154 in AngⅡ-induced cardiac fibroblasts was upregulated.Transfection with miR-154 inhibitor could downregulate miR-154 expression in AngⅡ-induced cardiac fibroblasts.The miR-154 inhibitor reduced the expression of β-catenin,c-myc,CoL-Ⅰ,CoL-Ⅲ,and α-SMA in Ang Ⅱ-induced cardiac fibroblasts.Wnt/β-catenin activator treatment could reverse the inhibitory effects of miR-154 downregulation on the expressions of CoL-Ⅰ,CoL-Ⅲ,and α-SMA in AngⅡ-induced cardiac fibroblasts.Conclusion Downregulation of miR-154 reduces collagen expression in Ang Ⅱ-induced cardiac fibroblasts by inhibiting Wnt/β-catenin signaling pathway.关键词
