Exploring differentially co-expressed genes between rheumatoid arthritis and acute myocardial infarction using bioinformatics and LASSO-Cox regression analysis

Abstract
Objective To explore the comorbid relationship between rheumatoid arthritis(RA)and acute myocardial infarction(AMI)using bioinformatics and LASSO-Cox regression analysis,and to identify potential biomarkers with clinical value.Methods Gene microarray datasets for RA and AMI were retrieved from the GEO database.Differential gene expression analysis was performed using the"limma"package in R,followed by screening of differentially co-expressed genes shared between the two diseases through weighted gene co-expression network analysis(WGCNA).Functional enrichment analyses of GO and KEGG were subsequently conducted on these genes.Subsequently,protein-protein interaction(PPI)network construction,LASSO regression,and multivariate Cox regression analysis were performed on the obtained differentially co-expressed genes to further screen for key genes.Finally,the diagnostic value of these key genes for both RA and AMI was evaluated using nomograms and ROC curves,with validation conducted in an independent dataset.Results A total of 132 potential differentially co-expressed genes associated with RA and AMI were identified.GO analysis showed that the differentially co-expressed genes were mainly enriched in the biological processes,including immune response regulation and activation.KEGG enrichment analysis showed that differentially co-expressed genes may function through the related signaling pathways,including phagosome and osteoclast differentiation.Four shared key genes between two diseases(CST3,DYNLL1,HNRNPU,QKI)were identified by combining LASSO regression and multivariate Cox regression.Nomogram and ROC curves demonstrated that the above key genes had good diagnostic value for RA and AMI.Conclusion There are shared key genes between RA and AMI.It provides new theoretical insights for exploring the potential biological markers and drug targets in patients with RA complicated by AMI.关键词
