Abstract

[Objective]The aim of the study was to screening effective strains antagonizing konjac soft rot disease,revealing the mechanism of strain A30 in antagonizing konjac soft rot disease.[Method]The inter-root soil of konjac was collected from konjac planting bases in Yunnan province,and a biocontrol strain with control effect on Pectobacterium aroidearum was screened by plate standoff method,named A30,and the strain was identified morphologically and molecularly.Inoculation test of konjac bulb tissues and potting tests of preventive effect were used to validate the preventive effect of the strain against konjac soft rot,and the biocontrol properties of the strain were evaluated,and the biocontrol effects on cabbage were verified through potting tests.The growth-promoting properties of the strains were determined and pot tests were used to assess their growth-promoting effects on cabbage.[Result]Strain A30 was identified as Bacillus licheniformis,which inhibited P.aroidea-rum with an inhibition circle diameter of(1.20±0.08)cm.Antagonism tests showed that in the konjac bulb tissue inoculation tests by adding the P.aroidearum cell suspension and A30 cell suspension ratio of 1∶1,1∶2 and 1∶3 mixture of 20 μL each,the konjac bulb tissue disease index compared with the inoculation of the P.aroidearum cell suspension alone were reduced sepacately by 6.96,14.90 and 23.57,which could effectively reduce the incidence of konjac soft rot,and the higher the concentration of antagonistic effects the higher the concentration,the better the antagonistic effects;With the addition of A30 cell suspension in the efficacy pot test,the disease index of konjac bulb tissue was reduced by 20.45 compared with that of inoculation with P.aroidearum suspension alone,and the relative efficacy of prevention reached 28.58%.The growth promotion tests showed that strain A30 had the ability of nitrogen fixation,iron carrier production and indole acetic acid(IAA)production,and the A30 bacterial suspension significantly promoted the growth of cabbage seedlings.[Conclusion]B.licheniformis A30 screened from the rhizosphere of konjac exhibits good antagonistic effect and growth-promoting characteristics,and it has the ability to prevent and control the soft rot disease of konjac as well as a certain growth-promoting potential.
