基于小鼠宫颈癌模型的HPV16 E6抗原肽负载树突状细胞治疗效果评价
Analysis of the therapeutic effect of HPV16 E6 antigen-peptide-loaded DC cells on a mu-rine cervical cancer model

Abstract
Objective:To establish a mouse model of cervical cancer and to investigate the tumor suppressive effects and immune response of dendritic cells(DCs)loaded with HPV16 E6 antigenic peptides.Methods:Healthy female C57BL/6 mice aged 6~8 weeks were selected to establish a cervical cancer model by subcutaneous injection of TC-1 cells.When the tumors reached 15mm in size,the mice were evenly divided into three groups:control group,DC group,and DC+Peptide group.The tumor area of each group was compared,and tumor growth curves were plotted.Lymphocytes were isolated from the spleens of the mice,and flow cytometry was used to detect the levels of Treg(CD4+CD25+Foxp3+)and IFN-γ expression in each group.Results:The tumor area and weight in the DC+Peptide group were significantly lower than those in the control group and the DC group.The expression of CD4+IFN-γ and CD8+IFN-γ in the spleen tissue of the mice in the DC+Peptide group was significantly higher than that in the con-trol group and the DC treatment group(P<0.05).The number of Treg in the DC+Peptide group was significantly lower than that in the control group and the DC treatment group(P<0.05).Conclusion:DCs loaded with HPV16 E6 mutated antigenic peptides can effectively activate mouse T lymphocytes,trigger specific anti-tumor immune responses,and thus inhibit tumor growth.关键词
