Cloning and Expression Analysis of Phosphorus Metabolism-related Gene PmGDPD1 in Pinus massoniana

Abstract
GDPD1 gene is critical for the regulation of phospholipid metabolism in plants,which plays an important role in stress resistance of plants.PmGDPD1 gene was cloned and its expression under low phosphorus stress was examined,in order to provide a basis for the study of PmGDPD1 function.PmGD-PD1 gene was cloned by RT-PCR and bioinformatics analysis was performed through an online website.Subcellular localization was analyzed by the transient transformation system in tobacco.Expression of this gene in plant tissues under low phosphorus stress was determined by RT-qPCR.The longest coding region of PmGDPD1 gene is 1 083 bp in length,encoding 361 amino acids and containing a conserved GDPD do-main.PmGDPD1 was clustered with its homologs in tobacco,wheat,and rice.Subcellular localization ex-periments showed that the protein was mainly localized in the cell membrane and expressed in small amounts in the nucleus.RT-qPCR showed that the expression of PmGDPD1 gene was induced after 24,48 and 60 days of low phosphorus stress in P.massoniana seedlings,and the expression was induced in differ-ent tissues.PmGDPD1 gene,which mainly functions at cell membranes,responds to low phosphorus stress,providing a reference for further exploring the mechanism of low phosphorus stress response of P.massoniana.关键词
