Abstract

In order to verify the presence of Anaplasma phagocytophilum(APH)infection,and to characterize its genetic evolution in domestic cats in China,452 feline blood samples collected from veterinary hospitals across 11 regions in 7 provinces from December 2012 to December 2019 were retrospectively detected for APH nucleic acid using semi-nested PCR.Positive samples were confirmed through BLAST sequence alignment and phylogenetic tree construction.3 positive samples(3/452)were identified,which were collected during summer months,including 2 from Changchun City(July 2018)and 1 from Changsha City(June 2019).The sequence of positive samples was 96.4%-99.2%similar to that of APH reference strains(GenBank accession No.ON807566.1 and ON795112.1,isolated from ticks in Changsha in July and August 2022,respectively),indicating high homology.It was confirmed that,through phylogenetic analysis based on 16S rRNA sequences,the sequences of all positive samples clustered in the specific evolutionary branch of APH.In conclusion,APH had been present in domestic cats in China for years,and its prevalent strains had a stable phylogenetic structure.An important scientific evidence was provided for early warning and monitoring of tick-borne diseases as well as development of relevant control strategies in China.
