Preparation of the Oil Emulsion Inactivated Vaccine of Novel Goose Parvovirus in Ducks

Abstract
Novel goose parvovirus(NGPV)causes short beak and dwarfism syndrome(SBDS)in ducks,characterized by developmental delay,macroglossia,and shortened beaks.To effectively prevent and control the disease,the preparation of NGPV inactivated vaccine was carried out.The NGPV SDHZ0512 strain was propagated(in duck embryo fibroblast cells),inactivated and emulsified to prepare 3 batches of inactivated vaccines.Then safety,immunogenicity,challenge protection,and field trials were conducted.Results showed that vaccination of 7-day-old Cherry Valley ducks with the three vaccine batches(1.0 mL/bird,intramuscular injection)did not cause any local or systemic adverse reactions.Latex particle agglutination(LPA)antibodies with high titer(4.1 log2-4.4 log2)were induced by all the vaccines when 21-day-old ducks were immunized for 28 days;LPA antibody titer reached 6.8 log2 when 16-week-old ducks were immunized with vaccines of batch No.202403 for primary immunization for 28 days,was up to 10.1 log2 after secondary immunization,and remained 4.5 log2 at 50-week old.It was found that,by challenge trials on offspring of immunized breeder ducks,the offspring of 30-week-old ducks achieved effective weight gain protection and no virus shedding,while those of 40-week-old ducks achieved effective weight gain protection,the shedding protection decreased(2/10 shedding),those of 50-week-old ducks showed only partial weight gain protection and no virus shedding(10/10 shedding).Ducklings of immunized and non-immunized breeder ducks were simultaneously tested and compared on the infected farm,it was found that,for the two groups,survival rates of 98.2%-98.3%and 94.2%-95.4%,average body weights of 3.07-3.08 kg and 2.80-2.81 kg,and feed conversion ratios of 1.81-1.83 and 1.96-2.08 were observed,respectively,and typical symptoms of the disease were shown in some ducks of the latter group.In conclusion,the 3 batches of inactivated vaccines prepared by the study were safe and effective,and could induce immunized Cherry Valley ducks to produce durable antibody,effectively protecting commercial duck flocks.关键词
