高效液相色谱-串联质谱法测定大鼠血浆中加兰他敏的浓度
Simultaneous determination of galantamine concentration in rat plasma by high-performance liquid chromatography tandem mass spectrometry

Abstract
Objective To establish a high performance liquid chromatography-mass spectrometry(HPLC-MS/MS)method for the determination of galantamine in the plasma of SD rats.Method The plasma sample was precipitated with 100%dichloromethane protein and separated using Water XBridge Amide C18(150 mm ×4.6 mm,3.5 μm)chromatography column.The mobile phase was A:deionized water containing 0.1%formic acid and B:acetonitrile containing 0.1%formic acid.Gradient elution,flow rate of 0.8 mL·min-1;Injection volume:5 μL;Column temperature:40 ℃.Used electric spray ion source(ESI).The positive ion mode multiple reaction(MRM)monitoring mode detects ion pairs of m/z 288.2→213.0(galantamine)and 294.2 → 216.1(galantamine-d6),respectively.The specificity of the method,the standard curve and the lower limit of quantification(LLOQ),the precision and recovery,the matrix effect,and the stability of the method were investigated.Result Galantamine showed good linearity within the range of 0.5~500 ng·mL-1,the standard curve was y=1.34×10-2x-1.81 ×10-3(r=0.998),and the LLOQ was 0.5 ng·mL-1,respectively.The accuracy within and between batches was within the range of 95%-115%,and the precision was less than 15%.The plasma matrix had no interference with the determination,and the sample stability was good.Applying this method to the pharmacokinetic study of galantamine in rat plasma,the main pharmacokinetic parameters:Cmax was(1 316.83±656.06)ng·mL-1,tmax was(0.42±0.13)h,t1/2 was(2.56±0.73)h,AUC0-t was(3 317.50±1 256.79)ng·h·mL-1,AUC0-inf were(3 340.00±1 245.52)ng·h·mL-1.Conclusion This method meets the requirements of biological sample analysis,is simple,efficient,and accurate,and is suitable for determining the concentration of galantamine in rat plasma and studying its pharmacokinetics.关键词
