摘要

Abstract

AIM A method was developed for the determination of 1-deoxynojirimycin in mulberry twig alkaloids tablets by ultra-high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).METHODS The sample solution was separated using a Shimadzu GIST HP AQ C18 column(100 mm×2.1 mm,3 μm),employing acetonitrile with 0.1%formic acid aqueous solution(10:90)as the mobile phase.The column temperature was maintained at 30 ℃ with a flow rate of 0.2 mL·min-1.Electrospray ionization(ESI)positive ions source and multiple-reaction monitoring modes were applied to analyze with the m/z 164.1→69.1 as quantitative ions for quantitative analysis.RESULTS There was a strong linear relationship between the peak areas and the concentration of 1-deoxynojirimycin,ranging from 0.503 to 25.150 μg·mL-1.The correlation coefficient was 0.999 3 and the spiking recovery was 100.66%with a relative standard deviation(RSD)of 2.33%.The RSDs for precision,repeatability and stability were all less than 4.0%.The mass fraction of 1-deoxynojirimycin ranged from 97.92%to 101.86%across 3 batches of samples.CONCLUSION The proposed method has good advantages of operating easily,highly sensitive,analyzing rapidly and good durability of column which can be used for the determination of 1-deoxynojirimycin in mulberry twig alkaloids tablets.
