定量系统模型DILIsym®在药物性肝损伤中的研究进展
Research progress of quantitative system model DILIsym® in drug-induced liver injury
摘要
Abstract
Drug-induced liver injury(DILI)is liver damage caused by chemicals,biologics,traditional herbal medicine or dietary supplements,which is a major problem in drug development and clinical practice.The quantitative system model DILIsym® aims to integrate physiologically based pharmacokinetic model,toxic mechanisms of DILI,and inter-individual variability of subjects to simulate changes in biomarker levels over time to predict the risk of DILI of compounds.The DILIsym® model has been applied to drug research in a variety of therapeutic fields,but its prediction accuracy is affected by factors such as individual biological variability.This review aims to explore the application of experimental data of in vitro hepatotoxicity mechanism in the construction of DILIsym® model,summarize the practical application of DILIsym® model in various treatment fields,and analyze the factors affecting its prediction accuracy,so as to provide important reference for drug development and clinical rational drug use.关键词
