黄连素调节HIF-1α/VEGF信号通路对烧伤大鼠创面愈合的影响
Effect of Berberine on Wound Healing in Burn Rats by Regulating the HIF-1α/VEGF Signaling Pathway

Abstract
Objective To investigate the effect of berberine on wound healing in burn rats by regulating the HIF-1α/VEGF signaling pathway.Methods Rats were randomly separated into CK group,Model group,berberine low-dose group,berberine high-dose group,and berberine high-dose+YC-1(HIF-1α inhibitor)group.The healing of rat wounds was observed and the wound healing rate was calculated.HE staining was applied to observe the morphology of wound tissue.ELISA method was applied to detect the levels of prostaglandin E2(PGE2),IL-10,IL-6,and IL-1β in wound tissues.Reagent kits were applied to detect content of MDA,activities of SOD and GSH-Px in wound tissue.Western blot was applied to detect the expression of HIF-1α,VEGF,VEGFR-2,MMP-2,MMP-9,and transforming growth factor-β1(TGF-β1)proteins in wound tissues.Results Compared with the CK group,the wounds of rats in the Model group were still not completely dry,with more obvious wounds,lower wound healing rates,and incomplete wound tissue structure,the levels of PGE2,IL-1β,IL-6,content of MDA,the expression of MMP-2,and MMP-9 proteins in the wound tissue were greatly increased,the levels of IL-10,activities of SOD and GSH-Px,and the expression levels of HIF-1α,VEGF,VEGFR-2,and TGF-β1 proteins in the wound tissue were greatly reduced(P<0.05).Compared with the Model group,the wounds in the berberine low-dose group and berberine high-dose group groups were basically intact,with a great increase in wound healing rate and a tendency towards normal tissue structure,the levels of PGE2,IL-1β,IL-6,content of MDA,the expression of MMP-2,and MMP-9 proteins in the wound tissue were greatly reduced,the levels of IL-10,activities of SOD and GSH-Px,and the expression levels of HIF-1α,VEGF,VEGFR-2,and TGF-β1 proteins in the wound tissue were greatly increased(P<0.05).YC-1 was able to alleviate the therapeutic effect of berberine on burn wounds in rats(P<0.05).Conclusion Berberine can reduce inflammation and oxidative stress in burn rats,promote burn wound healing,possibly be achieved by activating the HIF-1α/VEGF signaling pathway.关键词
