Rapid detection of malachite green residue in aquatic products by liquid chromatography tandem mass spectrometry

Abstract
A liquid chromatography-tandem mass spectrometry(LC-MS/MS)method was developed for the rapid quantification of malachite green(MG)and leucomalachite green(LMG)in aquatic products.Samples were rapidly extracted with acetonitrile and ammonium acetate as solvents.The extract was separated on a Thermo Hypersil GOLD C18 column(100 mm×2.1 mm,1.9 μm).A mobile phase of acetonitrile and 5 mmol/L ammonium acetate(0.1%formic acid,v/v)was employed in gradient elution.Detection was by electrospray ionization in positive mode with multiple reaction monitoring(MRM),and quantification used an internal standard.MG and LMG exhibited linear re-sponses over 0.125~2.500 μg/L,with correlation coefficients(R2)greater than 0.999.The limits of detection(LOD)and quantification(LOQ)were 0.25 μg/kg and 1.00 μg/kg,respectively.The recovery rates of sample ranged from 80.9%to 98.9%,with relative standard deviations(RSDs)of 0.9%to 5.2%(n=6).This method complied with current standards and demonstrated high throughput.It was suitable for rapid quantitative of MG and LMG residues in large-scale aquatic products screening.[Chinese Fishery Quality and Standards,2025,15(2):15-21].关键词
