植物乳杆菌Biohalo 12对小鼠过敏反应的抑制效果研究
Investigation of the anti-allergic properties of Lactobacillus plantarum Biohalo 12 in mice

Abstract
Objective To evaluate the anti-allergic effects of Lactobacillus plantarum Biohalo 12 in a murine model of systemic allergic reactions.Methods Twenty-four six-week-old female BALB/c mice with similar body weights were randomly assigned to three groups(n=8 per group):a blank group,a control group,and an experimental group.Sensitization in the control and experimental groups was induced by intraperitoneal injection of 2 μg of ovalbumin(OVA)on day 1,followed by booster injections of 6 μg OVA on days 14 and 28.The blank group received intraperitoneal injections of an equivalent volume of normal saline at the same time points.The experimental group were orally administered L.plantarum Biohalo 12 at a dose of 4×109 CFU per day,while the blank and control groups received an equal volume of sterile distilled water.On days 1,14,and 28,blood samples were collected from the tail vein to measure serum levels of OVA-specific antibodies,including IgE,IgG1,and IgG2a,to assess allergic reactions.On day 35,at the end of the experiment,body weights and liver-to-body and kidney-to-body weight ratios were recorded.Blood samples were collected via orbital bleeding for further assessment of serum OVA-specific antibody levels.Bronchoalveolar lavage fluid was collected to determine interleukin(IL)-4 and IL-10 concentrations via ELISA.Additionally,spleen cells isolated from the mice were cultured in vitro for 48 hours,and the levels of interferon-γ(IFN-γ)were detected.Results At the end of the experiment,there were no significant differences in body weight,liver-to-body ratio,and kidney-to-body ratio between the experimental group and the blank or control groups.However,the experimental group exhibited a significant reduction in serum concentrations of OVA-specific IgE and IgG1,along with a marked increase in IgG2a levels(all P<0.001).Additionally,concentrations of IL-4 and IL-10 in bronchoalveolar lavage fluid were significantly decreased(P<0.001 for both).Moreover,under OVA-stimulated in vitro culture conditions,the level of IFN-γ was significantly elevated in the experimental group(P<0.001).Conclusions Lactobacillus plantarum Biohalo 12 demonstrates a favorable safety profile and shows potential as an anti-allergic probiotic.It promotes the secretion of IFN-γ and IgG2a while reducing levels of sensitization-associated antibodies(IgE and IgG1)and the pro-inflammatory cytokine IL-4,thereby indirectly upregulating the expression of the anti-inflammatory cytokine IL-10.These findings suggest its potential utility in allergy prevention or treatment;however,further studies are needed to elucidate its precise mechanisms of action.关键词
