386例胃癌患者错配修复蛋白表达与临床病理特征及预后的相关性分析
Correlation analysis of mismatch repair protein expression with clinicopathological features and prognosis in 386 gastric cancer patients

Abstract
Objective To investigate the relationship between mismatch repair(MMR)protein expression,deficient mismatch repair(dMMR),and clinicopathological characteristics as well as survival prognosis in gastric cancer patients.Methods Tissue samples from 386 patients who underwent radical gastrectomy for gastric cancer at Weifang People's Hospital between January and December 2018 were collected,and tissue microarrays were constructed.Immunohistochemistry(IHC)was used to detect the expression of MMR proteins,including MutL homolog 1(MLH1),post meiotic isolated increasing protein 2(PMS2),MutS homolog 2(MSH2),and MutS homolog 6(MSH6).The correlation between MMR protein expression and clinical characteristics was analyzed.Kaplan-Meier survival curves were plotted,and Cox regression analysis was performed.Results dMMR was observed in 43 cases(11.14%).The loss patterns of MLH1 and PMS2 demonstrated high concordance(φ=0.623).dMMR status was significantly associated with patient age,sex,tumor location,tumor diameter,and N stage.The overall survival rate was significantly higher in the dMMR group compared to the proficient MMR(pMMR)group(log-rank P=0.0125).Multivariate Cox regression analysis identified dMMR as an independent prognostic factor for predicting survival in gastric cancer patients.Conclusion dMMR is associated with several clinicopathological features in gastric cancer.Patients with dMMR exhibit a more favorable prognosis,and dMMR status serves as an independent prognostic factor for survival in gastric cancer.关键词
