Abstract

The objective was to explore the quality differences between the wild-simulated cultivation and the film-mulching cultivation of Notopterygium forbesii,and provide a basis for the ecological cultivation of Notopterygium forbesii.Plant morphology observation,trait observation,microscopic characteristic observation and high-performance liquid chromatography(HPLC)were adopted,combined with principal component analysis(PCA),to analyze the plant morphology,traits,microscopic characteristics of the transverse section of the root,and the contents of seven active components(chlorogenic acid,nodakenin,ferulic acid,notopterol,phenylethyl ferulate,isoimperatorin,falcarindiol)of Notopterygium forbesii under two cultivation modes.In terms of plant morphology,two-year-old plants under wild-simulated cultivation presented a single main stem without tillers,and the plant height was significantly lower than that of the film-mulching cultivation group.However,the plants over two years old under film-mulching cultivation had clustered stems.In terms of traits,three-year-old wild-simulated products were smaller in overall size with obvious main roots(1-2 pieces),uneven root sections with cracks,and 1 to 2 stem bases at the top of the rhizome.While three-year-old products under film-mulching cultivation were larger in size with unclear main roots,multiple fibrous roots,uneven root sections,large and numerous cracks,and 1 to 7 or more stem bases at the top of the rhizome.In terms of microscopy,the cork layer of three-year-old wild-simulated products was thicker,the cracks in the phloem were few and small,and the xylem was well-developed.For the three-year-old products under film-mulching cultivation,the cork layer was thin,the cracks in the phloem were numerous and large,and the xylem was narrow.In terms of chemical components,there were no significant differences in the contents of chlorogenic acid,ferulic acid,phenylethyl ferulate and falcarindiol between the wild-simulated products and the film-mulching cultivation products(P>0.05).However,the contents of nodakenin,isoimperatorin and notopterol in the products under film-mulching cultivation were significantly higher than those in the wild-simulated products(P<0.05).The comprehensive score of the principal components of the contents of the seven components showed that the comprehensive quality of the products under film-mulching cultivation was better than that of the wild-simulated products.It is concluded that both wild-simulated and film-mulching cultivation modes have significant effects on the plant morphology,microscopic characteristics,and active ingredient accumulation of Notopterygium forbesii.Film-mulching cultivation performs is better in the accumulation of coumarin components(decursin,isoimperatorin,and notopterol).Wild-simulated cultivation maintains the ecological adaptability of plant resistance structures by simulating native habitat stress.The combination of the two cultivation performs can provide theoretical support for the green transformation of modern Chinese herbal medicine agriculture.
