加味葛根芩连汤质量控制检测方法的建立
Establishment of quality control detection method for modified Gegen Qinlian decoction

Abstract
This study aims to establish the quality control method of modified Gegen Qinlian decoction through high-performance liquid chromatography(HPLC),facilitating the modernization research and industrial production of classic prescriptions.Referring to the"Chinese Veterinary Pharmacopoeia"(Part Ⅱ)and related literature,a Hypersil ODS2 C18 column(4.6 mm×250.0 mm,5 μm)was employed.Chromatographic conditions were optimized for different components to determine the contents of puerarin,baicalin,berberine hydrochloride,liquiritin,atractylenolide,atractylon,ergosterol,and verbascoside in modified Gegen Qinlian decoction.The results indicated that the peak shapes of puerarin,baicalin,berberine hydrochloride,and liquiritin were complete,with high resolution.The negative control showed no interference at the corresponding retention times,suggesting good specificity of this chromatographic method.The RSDs of both the precision and repeatability experiments were less than 2.0%(n=6),demonstrating good precision and repeatability of the chromatographic method.The peak areas and retention times of each sample were recorded.The concentrations of puerarin,baicalin,berberine hydrochloride,and liquiritin in modified Gegen Qinlian decoction were calculated to be 0.31 mg/mL,1.52 mg/mL,0.15 mg/mL,and 0.63 mg/mL,respectively,providing a quantitative basis for the quality uniformity control of the prescription.However,atractylenolide,atractylon,ergosterol,and verbascoside were not detected in the corresponding samples,which might be attributed to the low solubility of these components or drug interactions that lead to the inability of detecting sensitively by the established method.关键词
