鸭疫里氏杆菌分离鉴定及紫锥菊等中药体外抑菌试验
Isolation and identification of Riemerella anatipestifer and in vitro antibacterial test of traditional Chinese medicine such as Echinacea purpurea

Abstract
In order to screen for traditional Chinese medicine with antibacterial effects against Riemerella anatipestifer,samples were taken from suspected diseased ducks in Meizhou,Guangdong Province,and pathogenic bacteria were isolated.The isolated bacteria were identified by morphology,Gram staining characteristics,biochemical characteristics,and PCR,and their ompA gene sequences were analyzed.At the same time,the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of traditional Chinese medicine extracts such as Echinacea purpurea and Callicarpa nudiflora were determined against Riemerella anatipestifer.The results showed that four strains of Riemerella anatipestifer were isolated,and all isolates were able to grow as smooth gray white circular colonies on blood agar plates.Gram staining was negative,and biochemical identification results showed that none of the strains fermented multiple sugars,produced hydrogen sulfide,could not liquefy gelatin,and were positive for catalase.The PCR identification results showed that a target band with a size of 707 bp was obtained.The results of traditional Chinese medicine susceptibility tests showed that the MIC values of Echinacea purpurea,Callicarpa nudiflora,Isatis indigotica,Rhubarb,and Folium Isatidis against Riemerella anatipestifer were 15.60 mg/mL,and their MBC values of all except Rhubarb(62.50 mg/mL)were 31.25 mg/mL.The MIC values of Scutellaria baicalensis,Astragalus membranaceus,and Lonicera japonica were all 125.00 mg/mL,and their MBC values were all 250.00 mg/mL.It indicated that Echinacea purpurea,Callicarpa nudiflora,Isatis indigotica,Rhubarb,and Folium Isatidis have good antibacterial effects on Riemerella anatipestifer.关键词
