裂殖壶藻粉营养强化对雄性斑马鱼生殖能力的影响
Effects of Nutrient Enrichment of Schizochytrium Powder on Reproductive Capacity of Male Zebrafish

Abstract
[Objective]To investigate the effects of nutritional enrichment with Schizochytrium powder on the reproductive performance of male zebrafish(Danio rerio),and to provide an effective strategy for regulating the reproductive capacity of sexually mature fish.[Method]Schizochytrium powder was selected as an aquatic dietary fortifier.Aquatic feeds with Schizochytrium powder addition levels of 0,40,80,and 120 g/kg were prepared.Sexually mature male zebrafish were fed with these fortified feeds for 30 days.By measuring phenotypic indicators such as fish body mass,body length,organ indices,and sex hormones were measured,and the gene expression levels of the reproductive gene regulatory axis,the mechanism of dietary Schizochytrium powder enhances male fish reproductive capacity were explored.[Result]Phenotypic studies on zebrafish showed that the body length of male zebrafish in all Schizochytrium powder-supplemented groups were significantly increased compared with the control group(P<0.05),among which the 80 g/kg group showed the maximum increase in body length(6.48%).The hepatosomatic index(HSI)of male zebrafish in the 80 g/kg and 120 g/kg groups compared with the control group were significantly elevated,with the 80 g/kg group exhibiting a 13.94%increase(P<0.05).Additionally,the gonadosomatic index(GSI)of the 80 g/kg group was significantly higher than that of other groups(P<0.05),indicating improved gonadal development ability.Observations of gonadal tissue sections showed that the proportion of spermatocytes in male zebrafish of the 80 g/kg group was(67.37±3.11)%,which was significantly higher than that of the control group(59.59±4.42)%(P<0.05),suggesting an increased degree of gonadal maturation.Analysis of sex hormone levels demonstrated that the serum testosterone levels of male zebrafish in all Schizochytrium powder-supplemented groups were significantly increased(P<0.05),and the gonadal estradiol-2 levels in the 40 g/kg and 80 g/kg groups were significantly elevated(P<0.05),indicating optimized sex hormone levels.Gene studies indicated that dietary supplementation with Schizochytrium powder could upregulate the relative expression of gnrh3 and lhβ in the brain of male zebrafish,thereby promoting gonadotropin secretion.It also increased the relative expression levels of androgen receptor(ar)and steroid synthase(17β-hsd)in the gonads,as well as the relative expression levels of vtg1 and vtg2 in the liver,ultimately enhancing the reproductive capacity of male zebrafish.[Conclusion]Nutritional enrichment with Schizochytrium powder can enhance the reproductive capacity of male zebrafish,with the most significant effect when the addition amount of Schizochytrium powder is 80 g/kg.关键词
