摘要

Abstract

[Objective]To investigate the dynamic transcriptome responses in the head kidney of largemouth bass(Micropterus salmoides)immunized with the attenuated Nocardia seriolae strain LY20810 F110,and to provide a theoretical basis for elucidating the vaccine-induced immunoprotective mechanisms.[Methods]Healthy largemouth bass were divided into immunized(injected intraperitoneally with 1×10⁸ CFU/mL attenuated N.seriolae)and control(injected with physiological saline)groups.Head kidney tissues were collected at 7 and 28 days post-immunization for RNA sequencing.Differentially expressed genes(DEGs)were identified via DESeq2 and subjected to KEGG pathway enrichment analysis.At 28 days post-immunization,fish were challenged with a N.seriolae virulent strain,and relative percent survival(RPS)was calculated alongside histopathological damage assessment.[Results]The immunized group achieved an RPS of 75%,with no granulomatous lesions or tissue damage post-challenge.Transcriptome analysis showed that 7 606 and 3 225 DEGs were identified at 7 and 28 days post-immunization,respectively.KEGG enrichment analysis revealed upregulation of innate(FcεRI signaling,complement C3)and adaptive(B-cell receptor signaling,NF-κB)immune pathways,along with increased expression of immune-related genes(IGH,CD22,PIGR),while pro-inflammatory pathways(IL-17 signaling,necroptosis)were downregulated.[Conclusion]Attenuated N.seriolae LY20810 F110 offers significant immunoprotection to largemouth bass,likely by activating innate and adaptive immune responses to eliminate pathogens and suppress excessive inflammation.
