银翘散HPLC指纹图谱建立及其抗流感病毒活性谱效关系研究
Establishment of HPLC Fingerprint of Yinqiao Powder(银翘散)and Relationship between Spectrum and Effect of Its Activity against Influenza Virus

Abstract
Objective:The high-performance liquid chromatography(HPLC)fingerprint of Yinqiao Powder(银翘散)was established to study its antiviral activity and screen its main active components.Methods:The HPLC method was used to establish the fingerprints of 24 batches of Yinqiao Powder samples,and the Similarity Evaluation System of TCM Chromatographic Fingerprint(2012 edition)was used to evaluate the similarity and identify the common peaks.The quality difference of 24 batches of samples was analyzed by chemical pattern recognition.The anti-influenza activity of Yinqiao Powder was detected by substrate fluorescence detection,and the main active components were screened by grey correlation analysis and orthogonal partial least square analysis(OPLS-DA).Results:The similarity of fingerprints of 24 batches of Yin-qiao Powder samples ranged from 0.854 to 0.982,and a total of nine common peaks were identified,among which forsythoside A,arctiin,and chlorogenic acid might be the main representative components leading to the differences among the samples.The 24 batches of samples showed good NA inhibition activity in the concentration ranging from 1.17 L to 75.00 mg/mL,with S21 achieving the highest inhibitory rate of 99.57%and S18 the lowest inhibitory rate of 89.24%.The antiviral biological titers of 24 batches of samples ranged from 2 909.76 U/mg to 8 320.71 U/mg,with S22 showing the highest titer,which was 2.86 times that of S13,the sample with the lowest titer.The degree of corre-lation between the six components and the biological activity titers of Yinqiao Powder against NA was above 0.70.There were three compo-nents with positive regression coefficient and variable importance in the projection(VIP)values greater than 1,which were arctiin,chlorogen-ic acid,and arctigenin.Conclusion:The established fingerprint of Yinqiao Powder can be used for quality evaluation,and the optimized sub-strate fluorescence detection method can be used to rapidly detect its antiviral activity.The spectrum and effect correlation study preliminarily identified the main active components of Yinqiao Powder for antiviral effects,which laid a foundation for the study of its quality control.关键词
