摘要

Abstract

Objective:To study the effect of Xiaocao(小草)Pills on hemorheology and heart injury in rats with myocardial ischemia of Hanningxu-eyu(寒凝血瘀)syndrome.Methods:Rats were randomly divided into normal control group,model control group,10 mg/kg aspirin group,85 mg/kg compound Danshen Dripping Pill group,and 0.5,1.5,and 4.5 g/kg Xiaocao Pill groups.Rats were administrated with correspond-ing drugs.After 1 h of administration,except for the normal control group,rats in other groups were placed in 0-4℃cold stimulation for 5 min,and isoproterenol(5 mg/kg)was injected intraperitoneally,for seven days continuously.The body mass of rats in each group was measured at 12 h and 24 h after the last administration.The cardiac ultrasonic function,hemorheology level,thrombin level,creatine kinase isoenzyme-MB(CK-MB)activity,cardiac troponin-T(CTn-T),von Willebrand factor(VWF),platelet α-granule membrane protein 140(GMP-140),cyclic adenosine monophosphate(cAMP),and cyclic guanosine monophosphate(cGMP)were measured.Hematoxylin-Eosin(HE)stai-ning was used to observe the pathological changes in the cardiac muscle tissue of rats and calculate the score.Results:Compared with that in the normal control group,the rats'body mass(on the 3rd,5th,and 7th day)was reduced in the model control group,as well as the contents of cAMP and cGMP(P<0.05,P<0.01).The left ventricular ejection fraction(LVEF),left ventricular fractional shortening(LVFS),stroke vol-ume(SV),cardiac output(CO),hemorheological indexes,prothrombin time(PT),activated partial thromboplastin time(APTT),CK-MB,CTN-T,cardiac mass index,VWF,GMP-140,and fibrinogen(FIB)were increased(P<0.05,P<0.01).Compared with the model control group,Xiaocao Pills could significantly increase the body mass of rats(on the 3rd,5th,and 7th day)and the contents of cAMP and cGMP(P<0.05,P<0.01).The levels of LVEF,LVFS,SV,CO,hemorheological indexes,PT,APTT,CK-MB,CTN-T,cardiac mass index,VWF,GMP-140,and FIB were decreased(P<0.05,P<0.01).HE staining showed that the myocardial injury,inflammatory infiltration,and myo-cardial cell widening in Xiaocao Pill groups were obviously improved,and the pathological score was significantly reduced.Conclusion:Xiao-cao Pills can effectively regulate the cardiac function,hemorheology,and coagulation indexes of rats with myocardial ischemia of Hanningxueyu syndrome and play a role in improving blood viscosity and myocardial injury,and its mechanism may be related to regulating the levels of VWF,GMP-140,cAMP,and cGMP.
