藏药仁青常觉对H22荷瘤小鼠的抑瘤及免疫调节作用研究
Study on Antitumor and Immunomodulatory Effects of Tibetan Medicine RenQingChangJue(仁青常觉)on H22 Tumor-Bearing Mice

Abstract
Objective:To study the antitumor effect of Tibetan medicine RenQingChangJue(RQCJ)on H22 tumor-bearing mice and its effect on tumor immune micro-environment and explore its mechanism.Methods:A total of 40 Kunming mice were randomly divided into five groups to establish a transplanted tumor model of H22 hepatoma mice.The positive control group was given an intraperitoneal injection of cyclophospha-mide(0.025 g/kg,once every other day),and 0.3 g/kg and 0.075 g/kg of RQCJ were daily given by gavage.The normal control group and the model control group were given equal amounts of normal saline.The mice were weighed every two days,and their weight changes were ob-served.Then,mice were killed after 10 days of continuous administration,and the tumor inhibition rate,as well as thymus and spleen indexes,were calculated.Hematoxylin and Eosin(HE)staining was used to observe the pathological morphological changes of the mouse's tumor tis-sue,and flow cytometry was used to detect the apoptosis of the mouse's tumor tissue.The expression levels of tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ),and interleukin-10(IL-10)in serum were detected by enzyme-linked immunosorbent assay(ELISA).The apoptosis of spleen cells and the number and ratio of CD4+/CD8+cells in T lymphocyte cells were detected by flow cytometry.The mRNA expressions of Caspase3,Caspase9,and Bcl2/Bax in the mouse's tumor tissue were determined by q-PCR.The protein expressions of PIK3CA,AKT 1,cleaved caspase 3,and Bcl 2/Bax in the mouse's tumor tissue were detected by immunohistochemistry.Results:Compared with those in the normal control group,the thymus and spleen indexes,the levels of IFN-γ and TNF-α,and the number of CD4+cells in spleen T cells were sig-nificantly decreased(P<0.01)in the model control group,while the IL-10 level and the apoptosis rate of spleen cells were significantly in-creased(P<0.01).Compared with that in the model control group,the tumor inhibition rate was 42.1%in the 0.3 g/kg RQCJ group(P<0.05).The apoptosis rate of tumor tissue,thymus and spleen indexes,contents of IFN-γ and TNF-α,the ratio of CD4+/CD8+in spleen T cells,mRNA expressions of Caspase3 and Caspase9,and protein expression of CLEAVED CASPASE 3 were significantly increased(P<0.05,P<0.01).The IL-10 level,the apoptosis rate of spleen cells,mRNA expression of Bcl 2/Bax,and protein expressions of PIK3CA,AKT 1,and BCL 2/BAX were significantly down-regulated(P<0.05,P<0.01).Conclusion:RQCJ has an antitumor effect on H22 tumor-bearing mice.It can improve the ratio of CD4+/CD8+in spleen T cells,regulate Th 1/Th 2 cytokine balance and tumor immune function,and promote tumor cell apoptosis.The mechanism may be related to the regulation of phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT)signaling path-way.关键词
