基于代谢组学的湖南晒黄烟虎斑特征挥发性物质解析
Analysis of Characteristic Volatile Compounds in Tiger-Stripe Sun-Cured Yellow Tobacco from Hunan Based on Metabolomics

Abstract
Tiger spot is a dark brown-yellow patch formed in the upper leaves of sun-cured yellow tobacco in Ningxiang,Hunan Province during the coloring period.Its appearance is closely related to the quality of tobacco leaves.In order to ex-plore the formation mechanism of the unique quality characteristics of tiger spot tobacco,this study used static headspace-solid phase microextraction-gas chromatography-mass spectrometry(HS-SPME-GC-MS)to compare and analyze the volatile metabolites of tobacco leaves with tiger spot and non-tiger spot tobacco leaves.The results showed that a total of 359 volatile metabolites were identified from tobacco leaves,of which ketones accounted for a high proportion,reaching 25.23%.Compared with non-tiger-spot tobacco leaves,the contents of ketones,alcohols,esters,and phenols in tobacco leaves with tiger-spot are relatively higher,and the content of 79 volatile metabolites(benzoaldehyde,neophydiene,so-lanisone,furfural,etc.)is significantly increased.Tobacco leaves with tiger spots have 32 specific metabolites,including 3-methyl-3-buten-2-one,5-methylcyclohexa-1,3-diene,(E)-hex-4-ene-3-one,etc.,which are mainly enriched in 8 metabolic pathways,including amino acid metabolism,carbohydrate metabolism,lipid metabolism,biosynthesis of other se-condary metabolites,energy metabolism,metabolism of cofactors and vitamins,metabolism of other amino acids,and meta-bolic pathways of terpenoids and polyketones;However,only three specific volatile substances were found in non-tiger leaf tobacco,which were mainly enriched in lipid metabolism pathways.Comprehensive analysis shows that the content of cha-racteristic aroma substances(neophydiene,solanidone,etc.)with high content in tobacco leaves with tiger spots can be used as a key indicator to evaluate the quality of the curing process of sun-cured tobacco leaves,providing scientific basis for the production of high-quality tobacco leaves.关键词
