Abstract

Objective:To observe the effect of salvianolic acid B(SalB)on glucocorticoid-induced growth retardation in juvenile rats and to explore its underlying mechanisms.Methods:Fifty-four 4-week-old SD female rats were randomly divided into the following groups:a normal control group,a model control group,a positive drug group with a compound calcium supplement(0.5 g/kg calcium gluconate+250 U/kg vitamin D3),and three SalB intervention groups at dosages of 12.5,25,and 50 mg/kg,with 9 rats in each group.Except for the normal control group,the rats in the other groups were administered 7 mg/kg prednisone acetate to establish the model and then treated with the corresponding drugs for 12 weeks.The rats'body weight was measured weekly.At the end of the experiment,body length and limb long bone length were meas-ured.The biomechanical properties of the femur were assessed using a three-point bending test,and bone histomorphometry analysis was con-ducted on decalcified tibial sections stained for bone tissue morphology.Results:Compared with the normal control group,the model control group exhibited significant reductions in body weight,body length,and limb long bone length(P<0.01).The femoral biomechanical proper-ties,including elastic load,maximum load,fracture load,and stiffness coefficient,were significantly decreased(P<0.01).The tibial longitu-dinal fluorescent spacing was narrowed,and the longitudinal growth rate was significantly reduced(P<0.01).The primary spongiosa width was notably reduced,indicating decreased new bone formation(P<0.01).Growth plate chondrocytes were disordered and abnormally hyper-trophied,with a significant narrowing of the growth plate thickness and the chondrocyte proliferation zone(P<0.01).Compared with the mod-el control group,the 50 mg/kg SalB group showed significant increases in body weight,body length,limb long bone length,and humeral wet weight(P<0.05).The femoral biomechanical properties were significantly improved(P<0.05).Furthermore,the longitudinal growth rate of the tibia and the primary spongiosa width were significantly increased(P<0.01).The morphology of chondrocytes in the growth plate was re-stored to near-normal,and both the growth plate thickness and the chondrocyte proliferation zone were significantly increased(P<0.05).Conclusion:SalB can significantly improve glucocorticoid-induced growth retardation and enhance bone quality in juvenile rats.The mecha-nism may involve the promotion of endochondral osteogenesis in the growth plates.
