对羟基苯甲醛对脑缺血后神经祖细胞迁移的作用及机制研究
Effect and Mechanism of p-Hydroxybenzaldehyde on Migration of Neural Progenitor Cells after Cerebral Ischemia

Abstract
Objective:To investigate the role and mechanism of p-Hydroxybenzaldehyde in regulating reactive astrocytes(RAs)to promote the mi-gration of neural progenitor cells(NPCs)after cerebral ischemia.Methods:Astrocytes(Asts)from cortex and NPCs were isolated and cul-tured in neonatal 24-h SD rats.Oxygen glucose deprivation/recovery(OGD/R)replica of the in vitro cerebral ischemia model was used to promote the activation of Asts to RAs and co-culture them with NPCs in transwell,and crystal violet staining was used to observe the migration of NPCs.Western blot was used to detect the expressions of apoptotic factors BCL-2 and BAX in NPCs and erythropoietin(EPO)which pro-moted the proliferation and differentiation of NPCs,neural mucoprotein(NEUROCAN)which inhibited the migration of NPCs,and polysialic acid neural cell adhesion molecule(PSA-NCAM),which promoted the migration of NPCs in RAs.Brain-derived neurotrophic factor(BDNF)was detected by ELISA for RAs promoting the survival of NPCs.Results:Compared with those in the blank control group,the number of mi-grated NPCs was increased in the model control group(P<0.05),and the ratio of BCL-2/BAX was significantly decreased(P<0.05).The expression of NEUROCAN protein in RAs was significantly increased(P<0.01),and the expressions of EPO and PSA-NCAM were signifi-cantly increased(P<0.01).BDNF content was significantly elevated(P<0.05).The migration number of NPCs was significantly increased after treatment with p-Hydroxybenzaldehyde(P<0.05),and the BCL-2/BAX protein expression ratio was significantly elevated(P<0.05).The expression of NEUROCAN protein was significantly decreased in RAs(P<0.05),while the expressions of EPO and PSA-NCAM proteins were significantly increased(P<0.05).The BDNF expression was significantly elevated(P<0.05).Conclusion:p-Hydroxybenzaldehyde can increase the migration and survival of NPCs,and this effect may be related to reducing apoptotic factors produced by NPCs and increasing migration and expression of chemokines and neuroprotective factors produced by RAs.关键词
