Abstract
Objective:To study the uric acid(UA)-lowering effect of Yejuhua(野菊花)extract on hyperuricemia caused by Guoshi Gaolianghou-wei(过食膏粱厚味)in rats and investigate the mechanism based on the reabsorption/excretion of UA.Methods:Fat emulsion was used to establish a rat model of hyperuricemia caused by Guoshi Gaolianghouwei.Eighty rats were randomly assigned into normal control,model con-trol,allopurinol(10 mg/kg),benzbromarone(5 mg/kg),and Yejuhua extract(15,30,60,and 90 mg/kg)groups according to their serum UA levels,with 10 rats in each group.During the experiment,the levels of UA and creatinine(CR)in the serum and urine were measured.The levels of blood urea nitrogen(BUN),alanine aminotransferase(ALT),aspartate aminotransferase(AST),total cholesterol(TC),triglyc-eride(TG),high-density lipoprotein cholesterol(HDL-c),and low-density lipoprotein cholesterol(LDL-c)in the serum were measured.The urinary beta-2-microglobulin(β2-MG)level was measured by enzyme-linked immunosorbent assay.Hematoxylin-eosin staining was employed to observe the histomorphological changes of the liver and kidney tissue.The protein levels of uric acid transporter 1(URAT1),glucose trans-porter 9(GLUT9),organic anion transporter 1(OAT1),and ATP-binding cassette transporter G2(ABCG2)in the kidney tissue were deter-mined by immunohistochemistry and Western blot.Results:Compared with the normal control group,the model control group showed a reduc-tion in kidney index,an elevation in serum UA level,decreases in urinary levels of FEUA and β2-MG,and elevations in serum levels of TC,HDL-c,LDL-c,AST,and CR(P<0.05 or P<0.01).Additionally,the model control group showed liver cells with normal size and morpholo-gy,clear boundaries,well arranged hepatic cords,and no obvious pathological changes.The modeling led to up-regulated protein levels of URAT1 and GLUT9(P<0.05)and down-regulated protein levels of OAT1 and ABCG2 in the kidney tissue(P<0.01).Compared with the model control group,Yejuhua extract administrated at different doses lowered the serum levels of UA,CR,ALT,and AST,increased the frac-tion excretion of uric acid(FEUA),alleviated the pathological changes in the liver and kidney,improved the liver and kidney functions,re-duced the levels of TC and LDL-c in the serum,ameliorated the lipid mechanism disorder,down-regulated the protein levels of URAT1 and GLUT9,and up-regulated the protein levels of OAT1 and ABCG2 in the kidney tissue(P<0.05 or P<0.01).Conclusion:Yejuhua extract can significantly lower the level of UA by reducing the UA re-absorption and increasing the UA excretion of the kidney.关键词
