刺五加提取物通过影响小脑代谢组学治疗帕金森病的作用机制
Therapeutic Mechanism of Ciwujia(刺五加)Extract on Parkinson's Disease by Modulating Cerebellar Metabolomics

Abstract
Objective:To explore the potential mechanism of action of Ciwujia(刺五加)extract in treating Parkinson's disease(PD)via metabo-lomics.Methods:The mice were divided into a normal control group,a model control group,and a group treated with Ciwujia at a dose of 45.5 mg/kg.The neurobehavior of mice was observed through the pole climbing experiment,rotational behavior experiment,and loco-motor activity experiment.Western blot and quantitative reverse transcription-polymerase chain reaction(qRT-PCR)were used to detect the protein and mRNA expressions of alpha-synuclein(α-syn)and tyrosine hydroxylase(TH).The cerebellum of mice in the normal control group,model control group,and Ciwujia group were analyzed by ultra-high performance liquid chromatography-quadrupole time-of-flight mass spec-trometry(UHPLC-QTOF-MS)method.Potential key biomarkers obtained from principal component analysis were compared with the Human Metabolomics Database(HMDB)and the Kyoto Encyclopedia of Genes and Genomes Database(KEGG)to identify relevant metabolic path-ways.Results:Compared with that in the normal control group,the mouse motion was significantly decreased in the model control group(P<0.01).The mRNA and protein expressions of α-syn were significantly upregulated(P<0.01),while those of TH were significantly downreg-ulated(P<0.05).Compared with the model control group,the Ciwujia group showed a significant increase in the mouse motion(P<0.01),a marked downregulation in the mRNA and protein expressions of α-syn(P<0.01),and a notable upregulation in the mRNA and protein ex-pressions of TH(P<0.05).A total of 15 potential biomarkers were identified in the cerebellum samples of PD model mice,which involved two main metabolic pathways,namely sphingolipid metabolism and glycerophospholipid metabolism.Conclusion:Ciwujia extract can inhibit α-syn aggregation by regulating sphingolipid metabolism and glycerophospholipid metabolism,upregulate the TH expression,and improve mo-tion function and neurodegenerative pathology in PD patients.关键词
