摘要

Abstract

Objective:To investigate the neuroprotective mechanism of electroacupuncture(EA)intervention in rats with cerebral ischemia-reperfusion injury(CIRI)and conduct metabolomic analysis.Methods:Forty-eight rats were randomly divided into sham-operated group(Sham),model group(CIRI),and electroacupuncture group(EA),with 16 rats per group.Except for the Sham group,other groups were subjected to CIRI modeling using Longa's suture-occlusion method.The EA group received treatment at"Zusanli"(ST36,cathode)and"Neiguan"(PC6,anode)with dense-disperse waves(2/15 Hz,0.5 mA,30 min/session).Body weight changes were recorded.Neurological deficits were assessed using modified Neurological Severity Score(mNSS)at various time points.Cerebral infarction area was analyzed by 2,3,5-triphenyltetrazolium chloride(TTC)staining.Pathological changes were evaluated by Nissl staining.Untargeted metabolomics was performed using liquid chromatography-mass spectrometry(LC-MS)to identify differential metabolites.Results:Compared with Sham group,the CIRI group showed:significant body weight loss;increased mNSS scores at all time points(P<0.001);elevated cerebral infarction percentage(P<0.001);neuronal damage with shrinkage,dark staining,and reduced intact neurons in Nissl staining;and altered metabolite abundances,particularly in carbohydrate-related compounds.Versus CIRI group,the EA group exhibited:increased body weight by day 7;reduced mNSS scores(P<0.001);decreased infarction area(P<0.001);and improved neuronal morphology/structure with increased intact neurons.Conclusion:EA at PC6 and ST36 reduces neurological deficits,decreases cerebral infarction volume,and repairs neuronal damage in CIRI rats,potentially through regulating neuroprotective metabolites related to glucose metabolism.
