土鳖虫活性肽治疗高脂血症的效果及初步机制研究
Therapeutic Effect and Preliminary Mechanism of Active Polypeptides of Eupolyphaga sinensis Walker on Hyperlipidemia

Abstract
Objective:To systematically evaluate the lipid-lowering activity of the active polypeptides of Eupolyphaga sinensis Walker(APE)using a hyperlipidemia(HL)rat model and preliminarily explore its potential molecular mechanisms.Methods:Seventy-two SD rats were randomly divided into a blank group(n=12)and a modeling group(n=60).An HL rat model was established by feeding a high-fat diet.After successful modeling,the rats in the modeling group were randomly divided into a model group,a simvastatin group(5 mg/kg),a high-dose APE group(100 mg/kg),a medium-dose APE group(50 mg/kg),and a low-dose APE group(25 mg/kg),with 12 rats in each group.The corresponding drugs were administered by gavage for 3 weeks,during which the body weight and mental state of the rats were recorded.Plasma and liver tissues were collected to measure the liver index,plasma total cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL-C),and high-density lipoprotein(HDL-C)levels,as well as liver alanine aminotransferase(ALT),aspartate aminotransferase(AST),interleukin-6(IL-6),and interleukin-8(IL-8)activities.HE staining and Oil Red O staining were used to observe liver pathological changes and lipid accumulation.Western blot and quantitative real-time PCR(qRT-PCR)were employed to determine the expression of AMPK-α,PPAR-α genes and CPT-1A,CPT-1B mRNA.Results:Compared with the blank group,the model group exhibited lethargy,increased body weight,liver index,serum TC,TG,LDL-C,and liver ALT,AST,IL-6,and IL-8 levels(P<0.0001,P<0.001,P<0.01,P<0.05),while HDL-C levels were significantly decreased(P<0.0001).Compared with the model group,the simvastatin group and medium-and high-dose APE groups showed reduced body weight(P<0.05)and liver index(P<0.01),improved mental state,and significant improvements in serum TC,TG,LDL-C,HDL-C,and liver ALT,AST,IL-6,and IL-8 levels(P<0.001,P<0.01,P<0.05).Liver pathological section observations revealed that APE had a certain reparative effect on liver injury and could ameliorate hepatic lipid accumulation.Western blot and qRT-PCR results showed that,compared with the blank group,the model group exhibited downregulated expression of lipid metabolism-related enzymes or genes(AMPK,p-AMPK,PPAR-α,CPT-1,CAT,HTGL)(P<0.01,P<0.05).After treatment with simvastatin and high-dose APE,the expression of these related indicators improved(P<0.01,P<0.05).Conclusion:APE demonstrates good therapeutic effects on HL,improving hepatic lipid accumulation and ameliorating hyperlipidemia by upregulating the expression of lipid metabolism-related genes such as AMPK,p-AMPK,PPAR-α,CPT-1,CAT,and HTGL.关键词
