基于AMPK/SREBP-1c/ACC信号通路探讨电针改善NAFLD大鼠肝脂质沉积的作用机制

Abstract
Objective:To observe the effect of electrotherapy on serum triglycerides(TG),liver tissue TG,liver steatosis and AMP-dependent protein kinase(AMPK)/sterol element-binding protein(SREBP)1c/acetyl-CoA carboxylase(ACC)signaling pathway in rats with nonalcoholic fatty liver disease(NAFLD),and to explore the potential mechanism of electroacupuncture in the treatment of NAFLD.Methods:Male SD rats were randomly divided into a normal group and high-fat diet group,and after eight weeks of feeding,a rat model of non-alcoholic fatty liver disease was evaluated.The NAFLD rat model was randomly divided into three groups:model group,electroacupuncture group,and electroacupuncture+AMPK inhibitor group.The electroacupuncture group received electroacupuncture stimulation at the"Zusanli"and"Pishu","Sanyinjiao",and"Fenglong"acupoints,while the electroacupuncture+AMPK inhibitor group received intraperitoneal injection of AMPK inhibitor Compound C on the basis of electroacupuncture treatment.Normal and model group rats were fixed on the platform without acupuncture.A total of four weeks of intervention were conducted to observe the body weight,serum TG,FFA in the liver group,triglyceride TG content in liver tissue,liver tissue pathology(HE staining),and Western blot detection of p-AMPK α,SREBP-1c,p-ACC expression and activation.Results:Compared with the normal group,the model group rats showed a significant increase in body weight,liver TG content,and HE staining of liver tissue with obvious steatosis.Compared with the model group,the electroacupuncture group showed a decrease in body weight,serum TG,liver tissue FFA,and liver tissue TG.The expression of p-AMPK α in liver tissue increased,and the levels of SREBP-1c and p-ACC decreased.Conclusion:Electroacupuncture can reduce liver lipid deposition in rats with NAFLD through the AMPK/SREBP-1c/ACC signaling pathway.关键词
