远志散通过调控脂质代谢改善大鼠记忆障碍
Yuanzhi(远志)Powder Improves Memory Impairment in Rats by Regulating Lipid Metabolism

Abstract
Objective:To investigate the mechanism of Yuanzhi(远志)Powder in improving memory impairment(MI)using lipidomics technology.Methods:A total of 27 male SD rats were randomly divided into the normal control group,the model control group,and Yuanzhi Powder 13.34 g/kg group,with 9 rats in each group.Except for the normal control group,rats in the other groups were intraperitoneally injected with D-galactose(0.4 g/kg)and intragastrically administered aluminum chloride(0.2 g/kg)daily to establish the MI model for 60 days.Starting from day 31,rats in 13.34 g/kg Yuanzhi Powder group were given Yuanzhi Powder by gavage once a day.After the last administration,the effect of Yuanzhi Powder on learning and memory functions was evaluated using the dark avoidance test.The levels of acetylcholine(ACh)and malondialdehyde(MDA),and the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in the cortex were measured.Ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS)was used to analyze the lipid metabolites in the cortex of rats and the metabolic pathways involved in the effect of Yuanzhi Powder on improving MI.The mRNA expressions of phosphatidyl-serine decarboxylase(Pisd)and phosphatidylethanolamine N-methyltransferase(Pemt)were detected by quantitative real-time polymerase chain reaction(qRT-PCR).Results:Compared with the normal control group,rats in the model control group showed a significantly shorter latency(P<0.05),significantly increased number of errors(P<0.05),and a higher error response rate.The levels of ACh and SOD in the cortex were significantly reduced(P<0.01),the GSH-Px activity was significantly decreased(P<0.05),and the content of MDA was signifi-cantly increased(P<0.05).The expression level of Pisd mRNA in the cortex was significantly upregulated(P<0.01),while the expression level of Pemt mRNA was significantly downregulated(P<0.01).Compared with the model control group,rats in the 13.34 g/kg Yuanzhi Powder group showed a significantly prolonged latency(P<0.05),reduced number of errors,and decreased error response rate.The levels of ACh,SOD,and GSH-Px activity in the cortex were significantly increased(P<0.05),and the MDA content was significantly reduced(P<0.01).The expression level of Pisd mRNA was significantly downregulated(P<0.01),while the expression level of Pemt mRNA was signifi-cantly upregulated(P<0.05).Lipidomics analysis identified 13 lipid metabolites,mainly involving glycerophospholipids(GP),sphingolipids(SP),fatty acids(FA),and prenol lipids(PR).GP metabolism was identified as the main metabolic pathway.Conclusion:The mechanism of Yuanzhi Powder in improving MI may be related to the regulation of oxidative stress and the GP metabolic pathway.关键词
