六味地黄提取物调控Wnt/β-catenin信号通路改善2型糖尿病大鼠胰腺损伤
Liuwei Dihuang(六味地黄)Extract Relieves Pancreas Injury in the Rat Model of Type 2 Diabetes Mellitus via Regulating the Wnt/β-catenin Signaling Pathway

Abstract
Objective:To investigate the effect of Liuwei Dihuang(六味地黄,LD)extract on the expression of Wnt/β-catenin signaling pathway in the pancreas of the rat model of type 2 diabetes mellitus(T2DM)induced by streptozotocin(STZ)and decipher the possible mechanism of the extract in relieving the pancreas injury of T2DM rats.Methods:Thirty-six male SD rats were randomized into the normal control,the model,metformin(0.23 g/kg),and LD extract(3.4,6.8,and 13.5 g/kg)groups(n=6).Rats were fed with a high-fat diet and treated with a low dose of STZ for the modeling of T2DM.Rats were treated by gavage for 8 weeks,and then the body weight and food intake were observed.The levels of fasting blood glucose(FBG),insulin(INS),2 hour postprandial blood glucose(2 h PBG),total cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL-c),and high-density lipoprotein(HDL-c)were determined.The homeostasis model assessment-insulin resist-ance(HOMA-IR)was calculated.Hematoxylin-eosin staining was employed to observe the pathological changes in the pancreatic tissue.Western blot was employed to determine the protein levels of β-catenin,transcription factor 7-like 2(TCF7L2),and glycogen synthase kinase-3β(GSK-3β).RT-PCR was conducted to determine the mRNA levels of β-catenin,Tcf7l2,Gsk3β,and Gcg.Results:Compared with the nor-mal control group,rats in the model group showcased elevated levels of FBG,2 h PBG,TC,TG,and LDL-c,increased HOMA-IR(P<0.05),lowered levels of INS and HDL-c(P<0.05),and histopathological injuries in the pancreatic tissue.In addition,the modeling down-regulated the protein levels of β-catenin and TCF7L2 and up-regulated the protein level of GSK-3β(P<0.05),and it down-regulated the mRNA levels of β-catenin,Tcf7l2,and Gcg and up-regulated the mRNA level of Gsk3β(P<0.05)in the pancreatic tissue.Compared with the model group,LD extract at different doses lowered the levels of FBG,2 h PBG,TC,TG,and LDL-c,decreased HOMA-IR(P<0.05),elevated the levels of INS and HDL-c(P<0.05),and relieved the histopathological injuries in the pancreatic tissue.Furthermore,LD extract up-regulated the pro-tein levels of β-catenin and TCF7L2 and down-regulated the protein level of GSK-3β(P<0.05),and it up-regulated the mRNA levels of β-catenin,Tc7l2,and Gcg and down-regulated the mRNA level of Gsk3β(P<0.05)in the pancreatic tissue.Conclusion:LD extract relieves the glucose and lipid metabolism disorder,insulin resistance,and pancreas injuries in the rat model of T2DM by regulating the Wnt/β-catenin sig-naling pathway.关键词
