摘要

Abstract

Objective:To investigate the effect of the Tibetan medicine Zuowoniang(佐窝尼昂)on lipopolysaccharide(LPS)-induced acute lung injury(ALI)in mice.Methods:An total of 54 healthy male C57BL/6J mice were randomly divided into 6 groups:the normal control group,ALI model control group,Zuowoniang groups(0.615 g/kg,1.23 g/kg,and 2.46 g/kg),and dexamethasone acetate group(5 mg/kg),with 9 mice in each group.The mice in each group were intragastrically administered the corresponding drug or water once a day for a total of 7 days.1 h after the last administration,the mice in each group,except the normal control group,were injected with LPS(15 mg/kg,10 mL/kg)into the tail vein for modeling,and the mice in the normal control group were injected with an equal volume of physiological saline into the tail vein.The mice were sacrificed 24 h after modeling,and the lungs,spleens,and thymuses were weighed,the organ coefficients were calculated.The lung tissues were stained with hematoxylin-eosin(HE)to observe the pathological damage of the lungs,and the enzyme-linked immunosorbent assay(ELISA)was used to analyze tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6),and interleukin-1β(IL-1β)in serum and bronchoalveolar lavage fluid(BALF),fully automated hematology analyzer was utilized to measure the counts of white blood cells,neutrophils,and lymphocytes in BALF,as well as the percentages of neutrophils and lymphocytes.Real-time quantitative polymer-ase chain reaction(RT-qPCR)was used to detect the mRNA expression levels of extracellular regulated protein kinases(Erk),inhibitor kap-pa B α(Ikbα),IκB kinase α(Ikkα),myeloid differentiation factor 88(Myd88),nuclear transcription factor-κB p65(Nfκb p65),p38 mito-gen-activated protein kinase(p38 Mapk),and Toll-like receptor 4(Tlr4)in lung tissues.Results:Compared with those in the normal con-trol group,the lung index and thymus index of mice in the model control group were significantly increased(P<0.05),alveolar epithelial cell hyperplasia,interstitium accompanied by inflammatory cell infiltration,and pathological damage score of mice were significantly increased(P<0.05),TNF-α,IL-6,and IL-1β in serum and TNF-α in BALF were significantly increased(P<0.01),the neutrophil percentage was signifi-cantly decreased(P<0.05),while the count of white blood cells,the absolute value of neutrophils,the absolute value of lymphocytes,and the percentage of lymphocytes all increased,but the difference was not significant,Iκbα,Ikkα,Myd88,Nfκb p65,p38,and Tlr4 mRNA expression were significantly elevated,with a highly significant difference(P<0.01),Erk mRNA expression was elevated,and the difference was signifi-cant(P<0.05).Compared with the model control group,Zuowoniang significantly reduced the lung index of ALI model mice(P<0.01),as well as interstitial inflammatory cell infiltration,pulmonary edema,and lung histopathological damage score to different degrees,but the differ-ence was not significant,Zuowoniang(2.46 g/kg)significantly reduced the serum TNF-α and IL-1β in ALI model mice(P<0.05),and TNF-α in BALF was significantly reduced in the Zuowoniang groups(2.46 g/kg and 0.615 g/kg)(P<0.01),the percentage of neutrophils and the absolute value of lymphocytes were significantly increased in the Zuowoniang groups(2.46 g/kg and 1.23 g/kg)(P<0.01 or P<0.05).Zuowoniang(2.46 g/kg)greatly reduced the mRNA expressions of Erk,Ikkα,Iκbα,Myd88,Nfκb p65,p38,and Tlr4(P<0.01 or P<0.05),and Zuowoniang group(1.23 g/kg)significantly decreased the mRNA expressions of Iκbα,Myd88,and Tlr4(P<0.05).Conclu-sion:Zuowoniang improves LPS-induced ALI,and the specific mechanism may be related to its ability to reduce the infiltration of interstitial inflammatory cells and inhibit the activation of NF-κB and MAPK inflammatory signaling pathways and the release of inflammatory factors.
