摘要

Abstract

Objective:To observe the effect of Qibai Pingfei Capsules on airway remodeling in rats with chronic obstructive pulmonary disease(COPD)by regulating the high mobility group box 1(HMGB1)/Toll-like receptor 2(TLR2)/nuclear factor-κB(NF-κB)signaling pathway.Methods:A total of 60 male SD rats were randomly divided into five groups:the normal control group,the model group,500 mg/kg Qibai Pingfei Capsule group,100 mg/kg pyrrolidinedithiocarbamate ammonium(PDTC)group,and 500 mg/kg Qibai Pingfei Capsules+100 mg/kg PDTC group,with 12 rats in each group.The COPD rat model was established.After six weeks of drug intervention,the lung function indexes of rats were detected.The expression levels of interleukin-6(IL-6),tumor necrosis factor-ɑ(TNF-ɑ),and NF-κB were detected by ELISA.HE staining was used to observe the pathological changes in lung tissues and the thickness of the airway wall.The positive expression levels of HMGB1,TLR2,and NF-κB p65 protein in the lung tissues of rats were detected by immunohistochemistry(IHC).The distribution and ex-pression levels of HMGB1,TLR2,and NF-κB p65 in the lung tissues of rats in each group were observed by immunofluorescence staining(IF).The protein expression levels of HMGB1,TLR2,and NF-κB p65 in the lung tissues of rats were detected by Western blot.Results:Compared with the normal control group,in the model control group,the bronchial lumen was narrowed,and a large number of inflammatory cells were distributed inside.The smooth muscle and basement membrane in the wall were obviously proliferated,and the thickness of the bronchial wall was significantly increased.The lung function index was significantly decreased(P<0.01).The serum levels of NF-κB,IL-6,and TNF-ɑ were significantly increased(P<0.01).The protein expression levels of HMGB1,TLR2,and NF-κB p65 in lung tissues were sig-nificantly up-regulated(P<0.01).Compared with the model control group,the inflammatory cells in the bronchi of rats in each drug group were significantly reduced,and the proliferation of the basement membrane and smooth muscle in the wall was reduced.The serum contents of NF-κB,IL-6,and TNF-ɑ in each group were significantly decreased(P<0.01).The lung function index was significantly increased(P<0.01).The protein expression levels of HMGB1,TLR2,and NF-κB p65 in lung tissues were significantly down-regulated(P<0.01),and the effect of 500 mg/kg Qibai Pingfei Capsules+100 mg/kg PDTC was the most significant(P<0.01).Conclusion:Qibai Pingfei Capsules may improve COPD airway remodeling by inhibiting the HMGB1/TLR2/NF-κB signaling pathway and reducing the release of inflammatory factors in the airway such as IL-6,NF-κB,and TNF-ɑ.
