桑寄生总多糖干预肾阳虚、肾阴虚骨质疏松模型及其寒热的研究
Effects of Total Polysaccharide of Sangjisheng(桑寄生)on Osteoporosis with Shenyangxu(肾阳虚)and Shenyinxu(肾阴虚)Syndromes and Its Han/Re(寒热)Medicinal Properties

Abstract
Objective:To investigate the preventive and therapeutic effects of total polysaccharide of Sangjisheng(桑寄生)on osteoporosis in the ovariectomized rat models of Shenyangxu(肾阳虚)and Shenyinxu(肾阴虚)syndromes and evaluate its medical properties of Han/Re(寒热)by cytological methods.Methods:The ovariectomized rat models of osteoporosis with Shenyangxu and Shenyinxu syndromes were estab-lished by ovariectomy combined with intramuscular injection of hydrocortisone and gavage of thyroxine,respectively.The models were treated with the positive control medicine or total polysaccharide of Sangjisheng for 12 weeks.The general state of rats was observed,and the anal temperature,hot plate residence time,bone mineral density,pathological changes in the femur,and bone morphometric parameters were meas-ured.The biochemical indexes related to bone metabolism and energy metabolism in biological samples were detected by ELISA.Results:Compared with the ovariectomized rat model,the total polysaccharide of Sangjisheng increased the trabecular bone thickness(P<0.05),low-ered the serum levels of bone-specific alkaline phosphatase(BALP),bone gamma-carboxyglutamic acid-containing protein(BGP),tartrate-re-sistant acid phosphatase 5b(TRACP-5b),deoxypyridinoline(DPD),and type I collagen carboxy-terminal peptide(CTX-I),and elevated the level of estradiol(E2)(P<0.01).Compared with the ovariectomized rat model with Shenyangxu syndrome,the total polysaccharide of Sangjisheng increased the trabecular bone thickness(P<0.05),lowered the serum levels of BALP,BGP,TRACP-5b,and DPD,elevated the levels of procollagen I N-terminal propeptide(PⅠNP),calcium(Ca),phosphorus(P),triiodothyronine(T3),thyroxine(T4),adrenocortico-tropic hormone(ACTH),corticosterone(CORT),and increased the cAMP/cGMP ratio(P<0.01)by elevating the plasma cyclic adenosine monophosphorus(cAMP)level and reducing the cyclic guanosine monophosphate(cGMP)level(P<0.05).Compared with the Shenyinxu model group,the total polysaccharide of Sangjisheng increased the bone trabecular area percentage,bone trabecular thickness,and E2 level,lowered the serum levels of BALP,BGP,TRACP-5b,DPD,CTX-I,thyroid-stimulating hormone(TSH),T3,T4,ACTH,and CORT(P<0.01 or P<0.05),and decreased the cAMP/cGMP ratio by reducing the plasma cAMP level and increasing the cGMP level(P<0.01).The results of cellular experiments showed that the total polysaccharide of Sangjisheng demonstrated the medical property of Wenre(温热).Conclusion:The total polysaccharide of Sangjisheng exerts the anti-osteoporosis effect by regulating Shenyangxu or Shenyinxu syndrome,with the medical property of Wen/Re.关键词
