The Effect of Acupuncture on Pain Behavior and Pain Inflammatory Factors in Rats with Sciatic Nerve Pain

Abstract
Objective:To observe the effect of acupuncture on pain indexes in sciatica rats and explore the mechanism of acupuncture treatment in sciatica rats.Methods:A total of 36 adult male SD rats with SPF grade were randomly divided into 4 groups by random number table method,namely blank group,model group,acupuncture group and drug group,with 9 rats in each group.The blank group did not do any treatment,and the model group,drug group and acupuncture group were modeled by clamping sciatic nerve.The success of the model was judged by foot blotting and hot and cold pain detector.24 h after successful modeling,the intervention was made separately.The blank group is not processed;The processing method of model group is the same as that of blank group.In the drug group,dezocine(2.5 mg·kg-1)was injected through muscle once a day,10 days a course,for 2 courses of treatment;In the acupuncture group,the rats were acupuncture at Huantiao point and Yanglingquan point,30 min/time,10 days/course of treatment,and 2 courses of treatment.After treatment,body weight comparison,rodent pain assessment scale(NRS)score,sciatic nerve func-tion index(SFI),sciatic nerve tissue morphology were compared by HE staining,and the expression levels of CX3CL1 and TNF-α were compared by immunohistochemistry.Results:After intervention,the body weight of rats in blank group,model group and drug group increased,but the body weight of rats in acupuncture group decreased.Compared with the drug group of the same period,the body weight variation coefficient of acupuncture group had significant difference(P<0.01),but there was no significant difference between normal group and model group(P>0.05).Before intervention,NRS scores in model group,acupuncture group and drug group were higher than those in blank group.After intervention,NRS score in acupuncture group and drug group decreased com-pared with before intervention.The variation coefficient of NRS in acupuncture group was significantly different from that in drug group(P<0.01).Before intervention,SFI values of model group,acupuncture group and drug group were lower than those of blank group.After intervention,SFI values of model group,acupuncture group and drug group were increased to different degrees.There was no significant difference in intervention coefficient among all groups(P>0.05).HE results indicated that sciatic nerve injury was more obvious in the model group than in the blank group.At the same time,acupuncture and drugs can promote local neovascular-ization of sciatic nerve,but acupuncture has more obvious effect.The immunohistochemical results of sciatic nerve indicated that the expressions of CX3CL1 and TNF-α in sciatic nerve pain model were higher than those in blank group,and the expressions of CX3CL1 and TNF-α in acupuncture group and drug group were lower than those in model group.Conclusion:Acupuncture and drug therapy have little difference in effect,both can reduce sciatica symptoms in rats,but acupuncture can increase the local blood sup-ply of sciatic nerve and promote the repair and metabolism of injured nerve.关键词
