电针预处理对大鼠急性肺损伤炎症反应及TLR4、NF-κB表达的影响
The Effect of Electroacupuncture Pretreatment on Inflammatory Response and Expression of TLR4 and NF-κB in Rats with Acute Lung Injury

Abstract
Objective:To observe the effects of electroacupuncture on the inflammation of rats with acute lung injury and the expres-sion of TLR4,NF-κB signal.Methods:Twenty-four male SD rats were randomLy divided into three groups:the control group,the acute lung injury group,and the electroacupuncture pretreatment group,eight rats for each.The acute lung injury model was estab-lished by Caudal vein injection of lipopolysaccharide.The rats in the electroacupuncture pretreatment group were given electroacu-puncture pretreatment before inject the LPS,electroacupuncture was used to stimulate bilateral Chize and Zusanli acupoints for 30min,once a day,for 5 days.The pathological changes in lung tissue of rats with acute lung injury were observed by HE staining,and evaluated by lung injury score.Serum levels of inflammatory factors IL-6 and TNF-α were detected by ELISA,mRNA and pro-tein expressions of NF-κB and TLR4 were detected by RT-PCR and Western-blot,respectively.Results:Compared with the control group,HE staining showed that the alveolar structures of rats in the acute lung injury group was destroyed and fused,lung injury score was significantly increased(P<0.01).Serum levels of inflammatory factors TNF-α were increased(P<0.01),IL-6 was also in-creased but the difference was not statistically significant(P>0.05).mRNA and protein expressions of NF-κB were significantly in-creased(P<0.01,P<0.01),mRNA and protein expressions of TLR4 were also significantly increased(P<0.01,P<0.01).Compared with the acute lung injury group,the damage degree of lung tissue structure in the rats of the electroacupuncture pretreatment group was significantly reduced,there was mild congestion and the degree of neutrophil aggregation was lower than that in the acute lung injury group,lung injury score reduced(P<0.01).Serum levels of inflammatory factors IL-6 and TNF-α were decreased,but the dif-ference was not statistically significant(P>0.05,P>0.05).mRNA and protein expressions of NF-κB were significantly decreased(P<0.01,P<0.05),mRNA and protein expressions of TLR4 were significantly decreased(P<0.01,P<0.05).Conclusion:Electroacupunc-ture reduce the inflammatory reaction by down-regulating the expression of TLR4 and NF-κB,and suppress the acute lung injury in-duced by lipopolysaccharide.关键词
