针刺联合自拟外敷贴治疗膝骨性关节炎模型大鼠的机制研究
Mechanistic Study on the Treatment of Knee Osteoarthritis Model Rats with Acupuncture Combined with Self-Formulated External Patches

Abstract
Objective:To observe the mechanism of acupuncture combined with self-formulated external patches in the treatment of knee osteoarthritis model rats.Methods:24 SPF SD rats were randomly divided into sham operation group,model group,acupunc-ture combined with self-formulated external patches group and western medicine control group,with 6 rats in each group.The knee osteoarthritis model was established by 4%papain saline in the model group,acupuncture combined with self-formulated external patches group and western medicine control group.The sham operation group was injected with the same amount of saline.After successful modeling,the corresponding intervention measures were given.The sham operation group was given normal saline by ga-vage.The model group did not take other intervention measures after successful modeling.The western medicine control group was given celecoxib capsule,and the acupuncture combined with self-formulated external patches group was given acupuncture com-bined with self-formulated external patches.Each group was continuously intervened for 14 days.The behavioral changes,joint function scores,expression of serum inflammatory factors and expression of related proteins in rats were compared before treatment and on the 1st,3rd,5th,7th,10th and 14th day after treatment.Results:Compared with the model group,the mechanical pain thresh-old and thermal pain threshold of the western medicine group and the acupuncture combined with self-formulated external patches group were significantly increased after 7 days of treatment,and the difference was statistically significant(P<0.05).Compared with the model group,the knee arthritis score of each administration group began to decrease after 5 days of treatment(P<0.05).Com-pared with the model group,the expression levels of MMP1,p65 total protein and nuclear p65 protein in the western medicine group and the acupuncture combined with self-formulated external patches group were decreased(P<0.05),and the expression level of nuclear p65 protein was increased(P<0.05).Compared with the model group,the expression levels of inflammatory factors TNF-α and IL-1β in the western medicine group and the acupuncture combined with self-formulated external patches group were decreased(P<0.05),and the expression level of IL-1β was decreased(P<0.05).Conclusion:Acupuncture combined with self-formulated exter-nal patches can improve the behavioral indexes and knee joint function of knee osteoarthritis model rats,reduce the inflammatory re-sponse,which may be related to the reduction of serum inflammatory factors IL-1β,TNF-α content and MMP1,p65 protein expres-sion levels.关键词
