摘要

Abstract

The rational design of enzymes has mainly focused on the active center and its surrounding area.However,in recent years,the research has gradually extended to other peripheral residues and even distal residues.The channels in enzymes,which connect the external solvent to the enzyme's active site,significantly influence catalytic activity,substrate specificity,and stereoselectivity of enzymes.Consequently,the peripheral amino residues constituting these channels have become a focus in rational design.The paper introduces methods for identifying and analyzing the channels in enzyme structures,reviews representative cases of enzyme channel modification,and explores the structure-function relationship between channel architecture and catalytic functionality in enzymes.
