摘要

Abstract

[Objective]Application of microbial agents is highly beneficial for improving soil and water environments.Characterized by enrichment of functional microbial communities,diversi-fication of microbial species,synergistic effects with natural flora,and safety in natural environ-ments,these agents possess the ability to degrade specific pollutants,thereby maintaining microbial dynamic balance.This study aimed to isolate and purify the microbial strains with high chromium re-sistance from three types of agricultural composite microbial agents for the remediation of chromi-um contaminated water and soil.[Method]Three microbial agents were selected.First,the control variable method combined with time dynamic monitoring was utilized to compare the removal rates of Cr(VI)in water by the three agents.Subsequently,the plate streaking method was employed to screen strains with high Cr(VI)resistance.The strains were preliminary identified by morphologi-cal and physiological-biochemical characteristics,and their species were confirmed via molecular biology.Growth conditions,including growth curves,optimal temperature,and pH,were investi-gated.The content of Cr(VI)was determined using the DPCI spectrophotometric method,and the removal rate of Cr(VI)by the strains was calculated.[Result]The experiment showed that 4 strains with good chromium resistance were screened from the 3 microbial agents,named G1,G2,S1,and T1.Strains G1 and G2 could tolerate a maximum Cr(VI)concentration of 200 mg/L,while S1 and T1 could withstand up to 550 mg/L and 500 mg/L of Cr(VI),respectively.Strains G1 and G2 belonged to Lysinibacillus,T1 belonged to Delftia,and S1 belonged to Microbacterium.The optimal pH for G1,G2,and S1 was approximately 7.0;S1 exhibited strong alkali resistance and grew well at pH 9.0,while T1 was acid-tolerant with an optimal growth pH of 6.0.The optimal growth temperature for G1 and G2 was 35℃,for T1 was 37℃,and for S1 was 40℃.The remov-al rates of Cr(VI)by the 4 chromium-resistant strains exceeded 99%in the chromium-containing culture solution with a Cr(VI)concentration of 100 mg/L.[Conclusion]The 4 screened microbial strains with good chromium resistance from 3 microbial agents possess the characteristics of high temperature resistance,alkali resistance,and rapid proliferation to occupy ecological niches.They could be applied to remediate the chromium pollution in water bodies and slightly saline-alkali soils in most ecosystems in China,making them promising strains for chromium pollution control agents.
