摘要

Abstract

Objective:To observe the antihypertensive and target organ protective effects of Tiaogan Huoxue Wenya Formula through animal experiments.Methods:A total of 24 13-week-old male SHR rats were randomly divided into 4 groups:the model group,the Tiaogan Huoxue Wenya Formula low-dose group(23.2 g/kg),the Tiaogan Huoxue Wenya Formula high-dose group(46.4 g/kg),and the amlodipine benzenesulfonate control group,with 6 rats in each group.A total of 6 13-week-old male WKY rats were set as the normal control group.The rats were treated for a total of 8 weeks with weekly measurement of systolic blood pressure(SBP),diastolic blood pressure(DBP),and mean blood pressure(MBP)in the tail artery;ELISA was used to deter-mine the level of angiotensinⅡ(Ang Ⅱ),aldosterone(ALD),angiotensin-converting enzyme 2(ACE2),endothelin 1(ET-1),high-sensitivity C-reactive protein(hs-CRP)in the serum of rats;H&E staining was used to observe the pathological changes in the heart,kidneys,and abdominal aorta of the rats,and Masson staining was used to observe the degree of fibrosis of abdominal aorta.Results:SBP,DBP,and MBP were significantly lower in both low-and high-dose Tiaogan Huoxue Wenya Formula groups compared to the model group(P<0.05),and those in the high-dose Tiaogan Huoxue Wenya Formula group were signifi-cantly lower compared to the low-dose group(P<0.05).The serum levels of AngⅡ,ALD,ET-1,and hs-CRP were significant-ly lower in both Tiaogan Huoxue Wenya Formula administration groups compared to the model group(P<0.05),and the ACE2 was elevated.Compared to the model group,the pathological degree of myocardial fiber arrangement disorder,fusion and fibrosis in the Tiaogan Huoxue Wenya Formula administration groups were improved,the renal tissue structure was restored,and the in-flammatory cell infiltration in the renal mesenchyme was improved.Compared to the model group,the myocardial fiber disorder in the abdominal aorta in the Tiaogan Huoxue Wenya Formula administration groups were improved,the lumen diameter was in-creased,the intima-media was thinned,the media thickness/luminal diameter was significantly reduced(P<0.05).The fibrosis of the abdominal aorta was improved,and the collagen volume was significantly reduced(P<0.05).Conclusion:Tiaogan Huoxue Wenya Formula has the ability to lower blood pressure and improve the injury of target organs such as heart,the kidneys,and blood vessels,and its pathway of action may be achieved by inhibiting the over-activation of the renin-angiotensin-aldosterone sys-tem,improving the function of vascular endothelium,and inhibiting the inflammatory response.
