复方茵陈蒿汤通过AMPK信号通路抑制皮脂腺细胞脂质合成和炎症因子分泌的机制研究
Mechanism of Fufang Yinchenhao Decoction in inhibiting sebaceous gland cell lipid synthesis and inflammatory cytokine secretion via the AMPK signaling pathway

Abstract
Objective:To investigate the effects of Fufang Yinchenhao Decoction on lipid synthesis and inflammatory cyto-kines in sebaceous gland cells mediated by the AMPK signaling pathway,and to elucidate its molecular mechanism in treating ac-ne.Methods:The human immortalized sebaceous gland cell line SZ95 was used to construct an acne cell model by inducting insu-lin-like growth factor-1(IGF-1).The Fufang Yinchenhao Decoction drug-containing serum or AMPK activator was subsequently administered.The effects on cell viability,apoptosis,and lipid droplet content in each group were assessed using the CCK8 assay,flow cytometry,and Oil Red O staining,respectively.Furthermore,the expression levels of lipid synthesis genes,inflammatory cytokines,and proteins related to the AMPK signaling pathway were quantified via qRT-PCR,ELISA,and Western blot,re-spectively.Results:Compared to the control group,IGF-1 significantly increased the viability of SZ95 cells,the area of Oil Red O staining,and the levels of IL-6 and IL-8,while inhibiting cell apoptosis.Additionally,it upregulated the mRNA expression of lipid synthesis genes SREBP-1,PPAR-γ,and LXR,as well as the expression of p-AMPK protein,and downregulated the ex-pression of NLRP3 and Caspase-1 protein(P<0.01).Compared to the model group,Fufang Yinchenhao Decoction significantly reduced the viability of SZ95 cells,the area of Oil Red O staining,and the levels of IL-6 and IL-8,while promoting cell apoptosis.Moreover,it upregulated the expression of NLRP3 and Caspase-1 proteins,downregulated the mRNA expression of lipid synthe-sis genes SREBP-1,PPAR-γ,and LXR,and suppressed the expression of p-AMPK protein(P<0.01).Conclusion:Fufang Yinchenhao Decoction exerts therapeutic effects on acne by activating the AMPK signaling pathway,and thus inhibiting lipid syn-thesis and inflammatory responses in sebaceous gland cells.关键词
