Abstract

Objective To explore the mechanism of Shenling Baizhu San regulating nuclear factor E2 related factor 2(Nrf2)/heme oxygenase-1(HO-1)signaling pathway in the intervention of ulcerative colitis,and to provide scientific basis for the treat-ment of ulcerative colitis(UC).Methods:Totally 72 SPF grade healthy male SD rats were selected and randomly divided into six different groups:the control group,the model group,the mesalazine treatment group(dose of 0.42 g/kg),as well as the low-dose group(0.54 g/kg),the medium dose group(1.08 g/kg),and the high-dose group(2.16 g/kg)of Shenling Baizhu San.Except for the control group,the UC model was established by free drinking of distilled water containing 5%dextran sodium sul-fate(DSS)in the other groups.After the molding,different concentrations of Shenling Baizhu San and mesalazine were used for gavage treatment for 14 days.The body weight,fecal character and blood in stool were recorded in each group of rats,the disease activity index was calculated,and the length of the rats'colon was measured;Hematoxylin-eosin staining was used to observe the histopathological changes in the colon of rats,and enzyme-linked immunosorbent assay was used to detect the levels of interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)in serum,as well as the content of superoxide dismutase(SOD)and malondialdehyde(MDA)in colon tissue;Western blot and RT-qPCR were used to detect the protein and mRNA expression of nuclear factor E2 related factor 2(Nrf2),heme oxygenase 1(HO-1),and quinone oxidoreductase 1(NQO1)in colon tissue.Results:Shenling Baizhu San can effectively relieve the symptoms of DSS induced UC rats,significantly reduce their Disease Ac-tivity Index score,and promote the recovery of colon length(P<0.01);the HE staining results showed that Shenling Baizhu San can reduce the formation of colonic mucosal ulcers,maintain the integrity of glandular structure,and reduce the infiltration of in-flammatory cells in the submucosal layer,further alleviating pathological damage to colonic tissue;Shenling Baizhu San not only increased the SOD content in colon tissue,but also reduced the levels of IL-1β,IL-6,TNF-α in rat serum and MDA in colon tis-sue(P<0.01);the results of RT-qPCR and Western Blot showed that the mRNA and protein expressions of Nrf2,HO-1 and NQO1 in colon tissues of rats in the treatment group were significantly increased(P<0.05),and the high-dose group of Shenling Baizhu San and the mesalazine group showed the most significant effect,with no statistical difference between the two groups(P>0.05).Conclusion:Shenling Baizhu San may inhibit oxidative stress response by activating the Nrf2/HO-1 signaling pathway,thereby playing a protective role in the colonic mucosa of UC rats.
