Abstract

Aim:To screen the potential biomarkers for gene diagnosis and immune infiltration of hepatocellular carci-noma(HCC).Methods:Gene expression profile microarray data of both normal adjacent(NAT)and HCC tissues from GEO database were used to screen the highly differentially expressed gene(HDEG)of HCC through LASSO regression and ran-dom forest model.Data from TCGA database were used to draw the ROC curve of gene diagnosis of HCC using HDEG,and lower limit of AUC 95%CI>0.8 was taken as the standard of identification of the potential biomarkers for gene diagnosis.The expression levels of target genes in 16 cases of HCC and paired NAT tissues from the Second Affiliated Hospital of Zhengzhou University were detected by qRT-PCR.The association between the target genes and 22 kinds of immune cells in-filtration characteristics were analyzed using single-sample gene set enrichment analysis algorithm based on GEO data.Re-sults:Apolipoprotein F(APOF),hepcidin antimicrobial peptide(HAMP),complement 9(C9)and metallothionein 1E(MT1E)were screened as potential gene diagnostic biomarkers for HCC,and the AUC(95%CI)were 0.953(0.933-0.973),0.946(0.922-0.970),0.926(0.901-0.951),0.925(0.899-0.950),respectively,and their expressions were all downregulated in HCC tissues compared with NAT(P<0.05).The infiltration levels of γδ T cells,monocytes,M2 macrophages,activated mast cells and neutrophils decreased,and those of activated memory CD4 T cells,regulatory T cells and MO macrophages increased in HCC tissues(P<0.05).APOF,HAMP,C9,and MT1E were correlated with the immune cells and function(P<0.05).Conclusion:APOF,HAMP,C9 and MT1E may serve as potential gene diagnostic biomarkers and therapeutic targets for HCC.
