蒙医温针对抑郁大鼠丘脑-垂体-肾上腺轴神经递质水平及神经元凋亡的影响
Impact of Mongolian medical warm acupuncture on hypothalamic-pituita-ry-adrenal axis neurotransmitter levels and apoptosis in depressed rats

Abstract
Aim:To explore the effects of Mongolian medical warm acupuncture(MMWA)on the hypothalamic-pituita-ry-adrenal(HPA)axis neurotransmitter levels and neuronal apoptosis of depressed rats.Methods:Forty rats were randomly divided into control group,model group,MMWA group,and MMWA+U1 group,with 10 rats in each group.The model group,MMWA group,and MMWA+U1 group were subjected to establish a chronic unpredictable mild stress model.MM-WA group received daily stressors for 1 hour followed by MMWA intervention,MMWA+U1 group underwent daily stressors and MMWA intervention,followed by intravenous injection of 10 μg/kg Urocortin 1(CRH receptor agonist),for 28 days.HE,Nissl and TUNEL staining were performed in the prefrontal cortex.ELISA was used to measure serum levels of adreno-corticotropic hormone(ACTH),cortisol(CORT),and prefrontal levels of 5-hydroxytryptamine(5-HT),norepinephrine(NE),acetylcholine(ACh),and dopamine(DA).The hypothalamic CRH mRNA levels were assessed using qRT-PCR,and prefrontal expressions of CRH,glucocorticoid receptor(GR),5-HT1AR,5-HT2AR,Bcl-2,and Bax proteins were detected by Western blot.Results:The model group exhibited significantly lower weight gain and sucrose preference rate,along with prolonged immobility time compared with the control group(P<0.05).Compared with the model group,the MMWA group showed increased weight gain,elevated sucrose preference rate,and reduced immobility time(P<0.05),while the MMWA+U1 group demonstrated no significant changes.Compared with the control group,the model group exhibited decreased protein expression of 5-HT,NE,ACh,DA,GR,5-HT1 AR,and Bcl-2(P<0.05);the number of apoptotic cells,serum levels of ACTH and CORT,hypothalamic CRH mRNA expression,and prefrontal protein expression levels of CRH,5-HT2AR,and Bax increased(P<0.05).Compared with the model group,the MMWA group showed improved levels in all these indicators(P<0.05),while the MMWA+U1 group did not show significant improvement.Conclusion:MMWA may exert its antide-pressant effect by regulating the HPA axis,thereby affecting neurotransmitter levels and prefrontal neuronal apoptosis.关键词
